Reaction of the sulfhydryl group of papain with chloroacetic acid.

چکیده
The kinetics of the alkylation of the essential sulfhydryl group of papain with chloroacetic acid was studied. The variation of the second order rate constant with pH at 4.5” and 30.5’, r/2 = 0.07, is bell-shaped, with rate constants at a maximum near neutrality, approaching zero at low pH, and approaching a positive minimum at high pH. These profiles are described by a formulation which relates the apparent rate constants both with the states of dissociation of the sulfhydryl and other groups in papain and with the absolute rate constants for sulfhydryl reaction under specific conditions of pH. This formulation, as well as the absolute rate constants and the dissociation constants, have been used to suggest the presence of groups near the active center sulfhydry1 group which affect the pH dependence of both the alkylation and the catalytic reactions of papain.

Microwave Assisted Synthesis of N-Benzylenaminones Catalyzed by Chloroacetic acid
Benzylaminoalkenones and benzylaminoalkenoates 3 were synthesized from reaction of benzylamine with corresponding 1,3-diketones and 1,3-ketoesters in the presence of catalytic amount of chloroacetic acid under microwave radiation. This method offers several advantages including high yield of products, recyclable of the catalyst and easy experimental work-up procedure.

Thiouracilis a historically relevant anti-thyroid preparation. Because of its structure you can find it in various chemical reactions differently. In this study, the reaction of Thiouracil with Chloroacetic acid and the formation of their additive products has been investigated. This reaction is aconcerted process, and it has not been determined yet by exhaustive mechanisms. From the potential ...

Facile and versatile procedures have been explored for the synthesis of tetrahydropyridines and 2,3-dihydroquinazolin-4(1H)-ones. These protocols employ the one-pot multi-component condensation of arylaldehydes and aromatic amines with β-keto esters and isatoic anhydride in chloroacetic acid, respectively. The reactions proceeded smoothly to generate the corresponding products in high yield. We...

متن کاملStudies on the active--SH group of papain and on the mechanism of papain activation by thiols.
It is now generally agreed that papain possesses a single reactive sulfhydryl group which is essential for activity and which probably participates in the catalytic mechanism by forming acyl intermediates with the different substrates (2, 3). Smith (4) has claimed that the essential sulfhydryl group of papain is kinetically more active than most protein sulfhydryl groups and that it possesses d...
